Basic Website Design

Hi All, I am looking to have a simple website developed using CSS, PHP and a MySQL db which I will be able to add graphical elements to at a later stage (I have some experience in site development). I would like to have the framework of the site completed through CSS. I do not have the site registered yet so I will need to be able to test the functionality by running it either locally or on a test site. Setting up the files on the future site is not required but some instructions on anything that will need to be done aside from transferring the files would be required. The main features to be implemented are a Twitter Integration and the ability to post News items on the site. I am looking to setup a Blog though I am planning on doing this by integrating the site with WordPress at a later stage I have attached a screen by screen mock up and some more detail around each feature.
